To acquire Lex Luthor’s Mech, you must have collected every Collectible: 195 Character Tokens (all but the Mech itself), 200 Gold Bricks, and 20 Red Bricks. Getting that means you’ll also have grabbed all 100 Minikits, all 20 Picture Perfect Moments (plus the 10 additional ones in the overworld) and all 15 Challenges.
